Dienogest Action Pathway

Dienogest is a semisynthetic progestogen derived from 19-nortestosterone and has antiandrogenic properties. This drug is used as a contraceptive or to treat endometriosis alone or combined with ethinylestradiol. Dienogest acts by binding as an agonist to the progesterone receptor with weak affinity but high specificity. This will lower the level of production of Gonadotropin hormone-releasing hormone (GnRH) in the hypothalamus. This results in the atrophy of the endometrium after prolonged use. As a contraceptive, it also acts by reducing the level of endogenous production of oestradiol, which suppresses the effects of oestradiol on both the eutopic and ectopic endometrium (anovulation). Also, since this drug is an agonist of the androgen receptor, it helps to improve androgenic symptoms like acne and hirsutism. This drug is administered as an oral tablet.
